What is Tirzepatide?

Tirzepatide is a synthetic peptide designed as a dual agonist of the GLP-1R and GIPR receptors. GIP (glucose-dependent insulinotropic polypeptide) is an incretin released in response to nutrients, acting on the pancreas, adipose tissue and nervous system. In SURMOUNT-1 (Jastreboff et al., NEJM 2022), the pharmaceutical formulation of Tirzepatide 15 mg weekly was associated with a mean 22.5% body-weight reduction at 72 weeks (treatment-regimen estimate), versus 2.5% in the placebo arm. This positioned Tirzepatide as the dual agonist with the strongest clinical evidence in weight management until the publication of Retatrutide data in 2023.
